I have a site with 2 WLAN; corporate and guest. On the Guest network, users are reporting not being able to update their exchange mail (outlook reportedly works fine).

So I added an extra rule to the Guest role to allow mail applications, but this still isn't working. Does exchange require additional rules?

Start off by allowing everything, then block what you do not want and then test.

Hmm... I guess :P Do I need to reboot the controller when making changes to Policies or are they enforced immediately?

No need for a reboot when updating ACL's, they will be enforced immediately. If you jump on the CLI and run #show datapath session | include XXXX (where XXXX is the IP your testing with) you can see any traffic being denied by locating  the D flag next to the traffic in question.
